PEGylated pH-sensitive liposomes (PSL) dual-loaded with gemcitabine and curcumin were investigated for the potential application in gemcitabine-resistant pancreatic ductal adenocarcinoma (PDAC) treatment. Curcumin was employed as an inhibitor of the efflux transporter, multidrug resistance protein 5 (MRP5) in PDAC cells. Liposomes were prepared with gemcitabine in the core and curcumin in the bilayers. The effects of curcumin on pH-sensitivity and 'endosome escape' of PSL with different PEGylation were investigated using a calcein self-quench assay. The effects of curcumin on intracellular gemcitabine concentrations, and cytotoxicity to a MIA PaCa-2 PDAC cell line was evaluated. The pharmacokinetics were investigated in rats following intravenous injection. The addition of curcumin to the PSL bilayers (0.2-1mol%)slightly decreased the pH-sensitivity of PSL, but to a less extent than PEGylation (0-5mol%). Co-treatment with curcumin increased gemcitabine cellular accumulation in a concentration-dependent ofiles for both drugs. While this is great news for individuals and their families, the traditional adult medical model must adapt to extend the care provided by specialty pediatric practices to primary care. In this paper, we introduce a model of integrated behavioral health (IBH) in a primary care practice for adults with childhood onset medical and developmental conditions. Our discussion includes the role of IBH providers (i.e., psychologists, psychiatrists, and social workers) as members of the integrated team, patient engagement and response to treatment, and innovative ways we strive to meet patient needs. Our review of electronic health records of patients seen at the UR Medicine Complex Care Center suggest that IBH is feasible and highly utilized, with 216 patients (40%) having had contact with an IBH provider on the team at least once. We discuss the challenges of meeting the longer-term needs of this complex patient population and our directions for future growth including creating peer and caregiver support networks, expanding services offered, and continued collaboration with community partners.In the field of drug-target interactions prediction, the majority of approaches formulated the problem as a simple binary classification task. These methods used binary drug-target interaction datasets to train their models. The prediction of drug-target interactions is inherently a regression problem and these interactions would be identified according to the binding affinity between drugs and targets. This paper deals the binary drug-target interactions and tries to identify the binary interactions based on the binding strength of a drug and its target. To this end, we propose a semi-supervised transfer learning approach to predict the binding affinity in a continuous spectrum for binary interactions. Due to the lack of training data with continuous binding affinity in the target domain, the proposed method makes use of the information available in other domains (i.e. source domain), via the transfer learning approach. The general framework of our algorithm is based on an objective function, which considers the performance in both source and target domains as well as the unlabeled data in the target domain via a regularization term.
