The use of selective leukotriene D4 receptor antagonists, targeting purinergic signaling as a therapeutic approach and glutamine supplementation may decrease the severity and mortality of COVID-19. A temporal relationship between hidradenitis suppurativa (HS) and obesity has not been established. To compare baseline body mass index (BMI) and change in BMI for patients with HS and controls before and after diagnosis. We performed a retrospective case-control analysis of 1284 patients with HS and controls matched for age, sex, race and calendar year between 1 January 1999 and 9 September 2019. BMI 7years prior to first HS diagnosis, and rate of BMI change, were compared for patients with HS and controls using linear mixed effects models. Baseline BMI was higher among patients with HS than controls [mean difference 3·03kgm , 95% confidence interval (CI) 2·44-3·62; P<0·001)]. This difference was larger for women [mean difference 3·37kgm (95% CI 2·73-4·01)] than men [mean difference 1·27kgm (95% CI-0·20-2·73; interaction P=0·01)], and for patients with HS diagnosed before age 40years [mean difference 4·10kgm (95% CI 3·15-5·06)] than those diagnosed after age 40years [mean difference 2·38kgm (95% CI 1·63-3·12; interaction P<0·001)]. Prior to diagnosis, average annual increase in BMI among patients with HS was slightly higher than controls. There was no statistically significant change in annual BMI among patients with HS following diagnosis. Baseline BMI and, to a lesser extent, rate of BMI change appear to be risk factors for developing HS. The influence of BMI may play a larger role among female patients and younger patients. Here, we identify a previously unrecognized subset within the CD127+ ILC population, characterized by the expression of the cytotoxic marker CD94. These CD94+ ILCs resemble conventional ILC3s in terms of phenotype, transcriptome, and cytokine production, but are highly cytotoxic. IL-15 was unable to induce differentiation of CD94+ ILCs toward mature NK cells. Instead, CD94+ ILCs retained RORγt, CD127 and CD200R1 expression and produced IL-22 in response to IL-15. Culturing non-cytotoxic ILC3s with IL-12 induced upregulation of CD94 and cytotoxic activity, effects that were not observed with IL-15 stimulation. Thus, human helper ILCs can acquire a cytotoxic program without differentiating into NK cells.We present the formulation, implementation, and performance evaluation of the Fourier pseudo-spectral method for performing fast and accurate simulations of electrophoresis. We demonstrate the applicability of this method for simulating a wide variety of electrophoretic processes such as capillary zone electrophoresis, transient-isotachophoresis, field amplified sample stacking, and oscillating electrolytes. Through these simulations, we show that the Fourier pseudo-spectral method yields accurate and stable solutions on coarser computational grids compared with other nondissipative spatial discretization schemes. Moreover, due to the use of coarser grids, the Fourier pseudo-spectral method requires lower computational time to achieve the same degree of accuracy. We have demonstrated the application of the Fourier pseudo-spectral method for simulating realistic electrophoresis problems with current densities as high as 5000 A/m2 with over tenfold speed-up compared to the commonly used second-order central difference scheme, to achieve a given degree of accuracy.
